Once the DTP goes live am I able to make any adjustments or updates to the novel? If so, do I have to wait another 48 hours for it to go live again?

I think once it goes live and you make adjustments it stays live till the new edition is finally adapted over, so it stays in circulation with no break. It rolls over the old upload

How long it takes for a change to go live seems to vary a lot. I recently helped a friend publish her book for Kindle. A few days later, we uploaded a new cover, which went live almost immediately. Around the same time, I uploaded a new book description for my own book, and it took about three days to go live.

However, pretend it's a print book and you have to get everything perfect first time. Otherwise it would be too tempting to publish it before it's really ready.

If you haven't already done so, I suggest you send the file to a few beta readers first, for feedback. I just enlisted the help of some from here earlier today.

I think it depends on what you change.  When I've changed the price or cover, it stayed live.  It took a day for it to change on Amazon, but it was available the whole time. When I changed the content, however, it was unavailable until the changes went through.  Not sure if that's true every time, as I've only ever done it once.  Maybe someone else has more experience with it.

I have made one change because the formatting was all messed up in the first one.  Luckily not many people had purchased the book   Sad

The revision went in over 48 hours and was seamless as far as I could tell.  Your buyers can request the revision at no charge by contacting Amazon's service department.

There have been a few times I have had to change things, and it always says my title will not be available for purchase for 24-48 hours, but my number of sales have always gone up DURING that time frame...

Get yourself an author's page on amazon.com and you can make a lot of the changes you need from there.

Be very careful with that "Live" notification.  Mine said it was "Live" but it wasn't yet priced and the product page didn't have a purchase button.  Thinking  I could change the product description without adversely affecting anything, I did just that.  My book then went back through both the "Review" and "Publishing" stages, setting final publication back by over a day and a half.  My book, which was in fact available for purchase on the U.K. site, also went back through both stages.

For the most part, the book will stay live.  If you upload a new cover or text file, it'll take a couple of days to take effect, but the old version will stay for sale until the moment the new book replaces.

EXCEPTION: Be careful when changing prices, especially when changing between 35% and 70% royalties, especially if your e-books are published elsewhere on the web.  This CAN cause your book to become unavailable in the Kindle store, sometimes for weeks.  Before messing with the price, make sure that you aren't underselling Amazon with a lower price elsewhere, or they will make your book unavailable for purchase.  If you distribute through a distribution site like Smashwords, also keep in mind that price changes can take weeks to take effect in all the bookstores.  So this is a long way of saying feel free to mess with anything but the price.

OK thanks all...good bit of info in a short amount of time. I don't need to change anything with the novel itself, just wanted to add a line at the bottom of the last page for my wife.

The DTP says the book is "publishing", however, my friend was able to buy it on Amazon.com already. I bought it myself to see what it looked like and I had it on my Kindle in a few seconds.
